I was looking for an antibody that can block the ligand binding domain of mouse EGFR. Can I use Cetuximab or is it exclusively human-specific?

No you cannot use cetuximab. There are slight differences between human and mouse epitopes. Cetuximab does not recognize mouze epitopes.

Looking at structure 1YY9 and comparing the UniProt sequences of human and mouse EGFR, 5 out of 7 contact residues in the huEGFR-Cetuximab complex are different in the mouse sequence.
No - these are sequences pulled from SwissProt/UniProt (www.expasy.org), which I aligned and colored according to similarity using a homemade EXCEL vba macro. I Identified contact residues using PyMOL in structure 1YY9 (www.rcsb.org) and marked the contact residues in the sequence alignment. The sequence difference in the epitope explains the lack of cross-reactivity mentioned in the paper cited in my first response. If you need something to cite for the sequence difference/epitope, you may use "A.Honegger, personal communication" ; )
