The application of bioleaching methods to the extraction of metals from minerals has definitely gained a prominent role and is supported by the several bioleaching plants operating in different places over the world. In recent years new techniques and new results about proteomic and bioinformatics are bringing a new perspective on the microbial processes.The psychrotolerant acidophile, Acidithiobacillus ferrivorans has been identified from cold environments and shown to use ferrous iron and inorganic sulfur compounds as the energy source.A bioinformatic evaluation suggests Acidithiobacillus ferrivorans utilized a similar ferrous iron oxidation pathway to the related species, Acidithiobacillus ferrooxidans. However, the inorganic sulfur oxidation pathway was less clear as the Acidithiobacillus ferrivorans genome contained genes assigned to encode for enzymes with functional redundancy from both Acidithiobacillus ferrooxidans and Acidithiobacillus caldus.

Acidithiobacillus caldus is an extremely acidophilic, moderately thermophilic, chemolithoautotrophic gammaproteobacterium that derives energy from the oxidation of sulfur and reduced inorganic sulfur compounds. Genome sequence of Acidithiobacillus caldus ATCC 51756 (the type strain of the species) have been carried out. This has permitted the prediction of genes for survival in extremely acidic environments, including genes for sulfur oxidation and nutrient assimilat.The A. caldus ATCC 51756 draft genome sequence has a total of 2,946,159 bp distributed in 139 contigs with an average GC content of 61.4%. Two 5S-16S-23S operons and 47 tRNAs on the draft assembly were identified, as were complete sets of genes for the synthesis of amino acids, nucleotides, and prosthetic groups.

With about 300 fully sequenced bacterial and archaeal genomes and with additional information of hundreds of thousands of DNA and protein sequences in public databases it is possible to predict genes and their putative protein products in DNA sequences derived from genome sequencing projects. In this chapter, it provides a general overview of how bioinformatics and genome biology can provide insight into the genomic organization and function of biomining microorganisms with a special reference to Acidithiobacillus ferrooxidans about which most is known. Bioinformatics and genome biology are effective tools for making preliminary inroads into how an otherwise uncharacterized organism functions. It is particularly powerful in cases where it is difficult to implement conventional genetic tools such as in the case of several bioleaching microorganisms and results are beginning to emerge to support this view
