I`m working on the tolerance of bacteria under drought stress applied using PEG6000 for future plant experiment.

I proceeded like:

1.Culturing the isolated bacteria (soybean soil) until the beginning of log phase

2.Checking OD at 600nm and diluting the OD to 0.1 (to have same cell density in all the strains)

3.Inoculating 1mL of the diluted bacterial strains in different concentrations of PEG (5%, 10%, 15%, 20% and 25%) diluted in TSB broth+ incubation for 24h

4.Checking turbidity and measuring OD (supernatant) after centrifugation of bacterial broth and compare it to the control (bacteria+TSB , no PEG)

However it seems like no one from the isolates tolerates the different concentrations of PEG

Am I missing something in my experiments?
