I am growing TG1 cells on 2XYT media without any antibiotic and getting two types of colonies, one is opaque and another one is transparent . i dont know which one is actually TG1. Can anyone help me ?

TG1 should be the opaque colonies. However, if the contaminant is consistently appearing when you plate from your stock then you may want to re-order the strain from the Yale collection.
I would also check morphology under a microscope.

They might both be TG1, frequently lon mutants of E. coli arise and those colonies will have a more mucoid/sticky/clear morphology. So it is hard to say if you have a contaminant or have picked up a mutant. Why not pick one of each type after streaking, grow up cultures and do a few easy phenotype screens.
