I am doing my assay on chitinase producing bacteria. I face the problem that I cant dissolve colloidal chitin in media in which I dissolve all the other ingredients in water.
I am working on chitinase producing microoganisms and I have used colloidal chitin agar for detecting chitinase activity of the organisms which produce a zone of clearance around its colony. For preparation of colloidal chitin agar, I incorporate 1% of colloidal chitin and agar into minimal salts medium.
Chitin can never be dissolved in any aqeuous solution. I presume you are preparing your own colloidal chitin. Just disperse it well in any suitable medium (with agar) and look for zone of clearence (if that is your sole purpose).
Dear all, thank you so much for your kind reply. I have done so and observed a clearing zone. but again around that hydrolyzed zone i could observe the growth of the bacteria again. does it happened with you also or there is something wrong in my experiment?
is there any differences between chitin and colloidal chitin. i'm also need to prepare the chitinase agar detection for my study but my lab only provided the chitin. All the formulations i found were using colloidal chitin. can i change to chitin? btw tq for your help
I am working on chitinase producing bacteria either,chitin doesn't dissolve in water. I assayed the NAGA produced in minimal medium plus 0.2% colloidal chitin but I am not able to culture them in the same solid medium. Can anybody help me for the reason?
Does colloidal chitin itself shows the reaction with Schale's reagent?
You need to make colloidal chitin and then disperse it in the agar medium having some nitrogen source like 0.5% peptone and may be 0.5% Na Cl and agar-agar 2.5%. Add colloidal chitin at 1% level.
I PREPARED COLLOIDAL CHITIN by dissolving 5gm chitin with 75 ml Hcl concentrated then stirred for one hour using magnetic stirrer then added 1000 ml distilled water . the supposed after that filtrated by whatmann No.1 filters in order to get residue but couldnt see the sediment in the bottom flask to filtrate if anybody prepares colloidal chitin how can i get residue or sediment from colloidal chitin ...best regards
