I have been working on extraction of mRNA of a gene of my interest in Arabidopsis. However, I am confused with what to expect in the gel after i run Total RNA (meaning mRNA, tRNA, rRNA) in a gel. Whats the best gel for Arabidpsis looks like and what are the sizesof th Band and what do they corresponds to ?
When you run your 1ug total RNA on the 1% agarose gel stained with ethidium bromide, you will see 2 prominent bands. These are the 28s and 18s rRNAs. The 28S bands is often thicker than the 18S, but both are distinctly clear.
The 28S corresponds to 4.8KB and 18S corresponds to 1.8KB when you use the DNA ladder as a marker.

Robert Pokoo , What makes you say that the rRNA component of Arabidopsis has 28S rRNA subunit and 18S rRNA subunit. I see that TAIR says the Arabidopsis has 25S rRNA and 18S rRNA
Hi Saugat , thanks for drawing my attention to it. The answer i provided was for total rna from most plants resolved on agarose gel.What you saw on the TAIR database is is right. Arabidopsis, Alternaria and Botrytis, just to mention a few show 25s and 18s when resolved on agarose gel.Most other plants commonly show the 28s and 18s bands.
