I have obtained a number of transgenic potato plants via overexpression of one of the genes from defensive pathways to enhance the resistance against the viruses. Then I evaluated transgenic plants using Real-Time PCR and ELISA. I want to know what morphological and physiological assessments can I do for our transgenic plants?

To assess their resistance to virus, you can artificially infect the transgenic and control plants to virus, and measure the growth rate through morphological (such as plant height, number of nodes, leaf area, fresh and dry weight, yield, ...) and physiological (such as chlorophyll, chlorophyll fluorescence, etc.) traits.

Evaluate for complete or partial resistance with refernce score for localised or systemic symptoms.
Please check the plants hormone contents especilly ABA and JA and compare them transgenic and control,if you want to check you need to investigate the diffrence between trichom size and number.these will be a strong evidence for the resistence and non resistance plants.
For industrial application yield also contribute one of the important parameters.
An evaluation of plant performance is a good way to assess the quality of the recovered plants. Yield in particular can be affected by passage through tissue culture.
In the first year you should artificially inoculate of the GM and the nontransformed plants or/and natural spread by the virus vector (aphids) and the experiment will monitored weekly by visual inspections. On the second and third years you will cultured the harvested tubers and determine both the decay of potato tubers and the stability of the resistance test.
