I am not a lipid raft biologist. My protein of interest accumulates in intracellular membrane puncta. We have data suggesting that our protein modulates PI3K/Akt/mTOR and PTEN, all of which can involve membrane microdomains called lipid rafts. So we are very interested in the precise nature of these membrane puncta that show vast enrichment of our protein of interest.

The most classical probe for lipid raft domains is based on the cholera toxin conjugated to a variety of fluors. These do not cross membranes.

We want to stain intact living cells, and we are sure that the membrane domains that interest us are within the cells and not exposed to the cell culture media.
