I want to do determine sGAG from the chondrocyte-alginate beads using a DMMB assay. In the literature, papain was used during the dissolution of the beads. So, I used papain, but did not manage to produce results. Perhaps I did not add the papain appropriately or at the right time. When and how much papain should I use in the DMMB assay? Can anyone provide any suggestions to improve my protocol or perhaps any other literature?
