I am growing 3D organoids from mouse lung AT2 cells in matrigel.
I am using glutamine, Penstrep. 10% FBS, ITS and tGF b inhibitor in media.after 6 days I observe large bubble like structure in the culture.
They appears like organoids but are huge in size in comparison to organoids.
How many percent of matrigel do you use? I guess the bubble-like structures that appeared on your samples are related to the matrigel.
I think it's completely normal if you get larger lung organoid like in your picture. As you can read in this review paper "Lung organoids: current uses and future promise (doi: 10.1242/dev.140103 )". You can read in the section of "organoids from mouse basal cells" where it stated " inhibitors of the BMP and TGFβ signaling pathways increased both CFE and cell proliferation (as assessed by EdU incorporation), resulting in larger diameter spheres with more cells ". Your matrigel looks fine.
