The lineweaver-Burk plot is also called Double-reciprocal plot. You need to convert the concentrations of substrate and the initial velocity into the reciprocals of them when using different concentrations of inhibitor. Attached is an example.  Thank you all for your suggestions. However I still face problem in plotting graph. Dr.Huang, the link was useful. However, I am not able to draw the trendline to determine if inhibitor is a competitive or non-competitive. It sounds like a silly question but still unable to figure it out. 

How many different concentration of inhibitors are you using? Did you mean you cannot get a focused point from different lines? or you did not obtain the linear regression with high R square? 

And also you probably fell victim to the effect of experimental error in double reciprocal plots.The lowest substrate concentrations may cause the greatest error, having more influence on the slope than the highest ones. You also may try using non-linear least squares regression to fit the data, such as MM equation

Hi Dr. Huang, as you mentioned, neither the points are getting focused nor am I able to obtain R2 value . So I cant determine clearly say the mode of inhibition. I have used two  inhibitor concentration. 

In my case lowest substrate concentration was not a problem. But the at the higher concentration there was large deviation.

You may go back to check the R2, the slop and the intercept of each line after you did linear regression. then you can calculate the intercept on the X-axis. You can see the line going through the minus value on the X-axis if the point (Intercept on the X-axis, 0) also is incorporated into the dataset. You may take a look at the example that I gave you. To determine the inhibition categories, it is better to have 3 different concentrations of inhibitor. It is more reliable. If this method does not work, you may try other method to determine the inhibitor classes. 

I believe that your problema is not how to make LB plots, but that you have used only two inhibitor concentrations. With only two points you cannot assure that the inhibitor is a linear (complete, full inhibitor) one and also to clearly distinguish among a competitive and a mixed inhibitor. If you are using Grahpad, you can test whether your experimental points fit better to a competitve inhibitor equation or two a mixed inhibitor. Search in your Graphpad versión.  Nevertheless, I insist that you should have at least three or better four inhibitor concentrations. Another way of analyzing the data you can find in my paper (Biochemistry International (1985) 11, 11-19) that is specialfor graphucally distinguish competitive from mixed inhibitors
