Hi, I am doing my macrophages culture from Porcine PBMCs of 3 months old piglets, So is it necessary that checking of maternally derived antibodies of a particular virus in that species? Again we have to give viral infection to macrophages after its complete growth and then harvest the cells. So please suggest me checking of MDA in piglets serum by cELISA is necessary?
Since you are isolating macrophages, culturing them and then infecting them in vitro, I do not think you need to assess the presence of MDA. Your macrophage cultures would be free of serum/circulating antibodies, thereby minimizing any impact. If you were doing in vivo infections, you would have to give some consideration to MDA (depending on how prevalent the virus is in your pig colonies and how long maternally derived antibodies stick around in weanlings).
thank you @ Julie Joseph for your reply, another clarification I need is that, If we isolated PBMCs from vaccinated animals/piglets blood for macrophages culture, is there any effect on macrophages from that virus which is used in vaccine?
and also is it interfere with macrophages in infection with the same virus?
It would depend on your vaccination/PBMC collection schedule and also how immunogenic your vaccine candidate is. If your vaccine is immunogenic and you are collecting your PBMCs once your animals have had time to generate a good virus-specific immune response, then the answer to both Q is yes. Your macrophages are probably going to respond to the virus in vitro and this could mean improved inflammatory responses. If your macrophage cultures are pure, you may not see killing but if you have T cells in there, they may be much better at killing the virus. All this would interfere with virus infection in vitro. However, this is not necessarily a bad thing esp. if you are looking for vaccine candidates.
