N. gonorrhoeae needs 5% CO2! A really easy and cheap way is to disolve 3 colonies in 150 µl aqua destillata and mix well in a tube (eppendorf). Then heat it at 95 °C in a thermoblock while shaking. After that put it in a utrasonic bath for 5 min. Now cetrifuge it at maximum and take the supernatant for PCR. Good luck!

Thanks for your reply, but the suggestion as you mentioned above is always my easy method that I use for screening with PCR amplification. What I encounter for now is I want to this genomic DNA for many important enzymatic reactions such as DNA sequencing, so I wonder that how can I grow those N. gonorrhoeae in liquid culture instead of picking colonies and dissolving them. 

Use Brain heart infusion broth supplemented with X and V factor and grow in 5% CO2.

Preferably with shaking and if that is impossible use a petridish with a thin layer of broth so the CO2 can saturate the medium. But still I think the bacterium will grow in lumps. But vortexing after transfering to a tube might do the trick.

N. gonorrohea is a fastidious bacterium, difficult to cultivate in liquid medium due to susceptibility to pH and divalent cation concentrations and a tendency to autolysis. Traditional broths like BRAIN HEART INFUSION may allow limited multiplication of N. gonorrhoeae, but typically require inocula in excess of 10^5 CFU mL^−1 to achieve dense growth consistently.

FB was prepared as described previously by Cartwright et al.
