After column chromatography I've got these two spots together. And tried a number of solvents ratio to increase the gap between these two spots for further column chromatography but failed.

instead of ethylacetate I would use solvents with different elution power.
List of solvents with different elution power:
Pentane / Hexane 0.00
Cyclohexane 0.04
CCl4 0.11
Toluene 0.27
Isopropyl ether 0.28
Chloroform 0.36
Diethylether 0.43
Ethyl acetate 0.48
Acetone 0.50
Acetonitrile 0.60

Methanol 0.95
Acetic acid >> 1

here I'm again. Standard solvent mixtures:
1. Toluene/ethyl acetate 95+5 for non-polar components
2. Chloroform/methanol/water 70+30+4 for saponins and lignans
3. Ethyl acetate/acetic acid/formic acid/water 100+11+11+27 for flavonoids
4. Acetonitrile/water/formic acid 30+8+2 for amino acids
5. Butanol/acetic acid/water 7+1+2 for polar components

Hii...first of all ... hope ur target compound is non-polar...u are targeting terpene category compounds ..or else...cause EtOAc hexane will not give u any spot or any separation between these compounds these are almost in nearer to the solvent front ... so better it's suggestible to increase polarity now ...like EtOAc to DCM ..then DCM MeOH ..then EtOAc to methanol best DCM chcl3 ...would be best you can spot those mid polar compounds ....and if possible fractionate first using VLC then use a column for each and every fraction.... by different gradient elutions ...available in eutrophic series ...
