I am using a methodology of pollen extraction from another in which anthers are shaken in hexane and pollen then extracted from hexane by millipore filter assembly. I want to replace the millipore filter assembly by simple whatman filter paper grade 5 (3 micron pore size). Upto my knowledge, size of mango pollen ranges between 10-15 micron. Can anybody has experience of such work? Any useful suggestions?

I have not worked with pollen, but just though that simple centrifugation would work? If you are passing your sample through a micron-sized filter that means that it is not very dirty with with big particles. You can precipitate bacteria with 4000xg in a centrifuge, so I suppose that something between 2000-6000xg would do for your pollen -as long as the density of the solution is lower than the density of the pollen.

Simple metal membrane holder (screwed) of required diameter and LuerLock inlet fiffing is the simplest way. You just cut (or buy) respective Whatman filters, put them (1 or 2) into the holder, screw it up and it is ready for use. It can be fitted to any syringe having the LuerLock fittings.
