I have been using the Thermo Scientific, Rockford, USA kit for IgM purification. During elution I don't feel like enough IgM is being eluted and its really troubling me. Any advice shall be highly appreciated.
I have eluted IgM from affinity chromatography monoclonal antibody attached to cyanogen-bromide sepharose 4-B, with best yields using ammonium thyocyanate, but also with 2 M NaCl, Tris-HCl, EDTA, or sodium sulfate.
You can use a ligand-affinity resin from LigaTrap Technologies. It really works for human IgM purification from hybridoma or 10%FBS cell culture media..
Binding capacity 5-10mg/mL resin..
It can recover up to 90% IgM from the total load with > 95% purity.
