I already have stained cells.
I am interested in relation to amount of fields and cells to be examined to make the relationship between normal cells and apoptotic.
Thanks in advance
you really should look at a minimum of 10 fields and 500 cells - if the rate of apoptosis is very low, you need to count more, especially if you are comparing different conditions.
Thanks!!
I daresay that 100 cells in five fields may be enough. You could check this article if you need further information.
Article Relationship between Caspase Activity and Apoptotic Markers ...
You can use flow cytometry method to quantify the positively stained apoptotic cells. The protocols are available with the TUNEL assay kits.
David, thanks for reply.
But I have a doubt, the fields don't show uniform distribution of apoptotic and normal cells, including, some fields only have "normal cells".
How do I choose the fields?

Im doing PBMC isolation -> CD14+ enrichment using magnetic beads -> stimulation setup. My negative control is just cells in cRPMI but they seem to get activated over and over again.
