I've been having trouble finding the right method for measuring interleukins in nasal wash samples. Some papers suggest centrifuging it, but I don't know if it works as doing it didn't leave a sobrenadant. The experts here say that you have to liquefy the mucus to measure them and some say that if you use a mucolitic it'll alter the results. What's the right protocol for this method?

The ELISA method is the best way to assaying cytokine. Briefly, the sample is homogenized in 2mL of ice-cold triple distilled water and sonicated for 15s. The homogenates will be then centrifuged at 10000 ×g for 2min and the supernatants are used for cytokine assay by the IL-1 ELISA kit.
