I am attempting to extract RNA from avian blood stored in RNAlater. Avian RBCs are nucleated, so the RBC lysis step (included in most protocols for RNA extraction from blood) is not needed. I have a PAXgene blood RNA kit (but samples are in RNAlater, not PAXgene tubes). I also have a QIAamp DNA blood mini kit. Does anyone know if I can hack some sort of useful RNA extraction from these kit components? I am trying to avoid buying a whole new kit because I need RNA from only 2-3 samples. Alternatively, does anyone have a non-kit RNA extraction protocol that I could use for avian blood (nucleated RBCs) in RNAlater?
After checking the PAXgene manual, it is likely the PAXgene RNA tubes use a reagent similar to RNAlater. After pelleting your cells and resuspending, you should be able to jump right back into the PAXgene protocol. If you have all of the components of the PAXgene kit, I would test the RNA purification with a sample you don't need or with half of one of your samples.
Be careful using DNA silica columns for RNA, the chemistry isn't always the same and you may lose your sample.
