I am in big trouble because of plasmid purification. I have several types of recombinant DH5alph which are stored in -70. Til 2 month ago I had no problem with them and manual plasmid purification were excellent but I don't know why when I add solution (potassium acetate) to it, it doesn't work ie the cloud does not form well and the Elastic form of it does not omit. I have checked my antibiotics and I have done colony PCR. Both of these test verified that the plasmid is present. I have changed the host. I used that bad plasmid for transforming new competent cells and again I have purified the plasmid but it was bad again. I have cultured them on solid LB. The appearance of the colonies were different. I did gerem staining it show no contamination. I checked my competent cells.They show no contamination. I have no isolation kit. I have check my buffers. Nearly no plasmid can seen on the gel. I have increased and decreased the early amount of bacteria. None of these test have helped me. In the past I could purified the same plasmid well. Now I cant.
