Ubiquitin acts chemotactic on monocytes via the SDF-1 receptor (CD184). To elucidate the role of ubiquitin for the influx of monocytes into wounds, we need an antibody to block ubiquitin in wound fluid. For our purposes it is important to prevent the binding of ubiquitin to this receptor. Every hint is highly appreciated. Thank you in advance.
the proteins involved in inflammation are all tightly regulated by post-translational polypeptide chain modifications, especially phosphorylation, ubiquitination and limited proteolysis.. in that case ubiquitin could be acting as a switch similar to excess substrate in the operon model.. as the inflammatory response reaches its climax, the proteins involved are sequestered for poly-ubiquitinantion.. this leads to to ubiquitin depletion within the cell, leading to detachment of the ubiquitin from the monocyte chemotactic protein-induced protein-1 (MCPIP1) complex ("link"), leading to monocyte non-entry and termination of inflammatory response.. this may be the possible role of ubiquitin during inflammation.. therefore, a GFP tagged real time video of ubiquitin may be employed to view changes in ubiquitin flux during inflammation and the fate of the receptor MCPIP1 bound ubiquitin!
