I am designing a delivery system using PEG-peptide conjugates. The peptide has the ability to self-assemble due to presence of a fatty acid or GF3 (tetra-mer). I dissolve the conjugates in nano pure water. I need help to interpret the TEM images. Images are attached.
1&2 show aggreggates like-fibers, laterally self-assembled, presence of clusters. Rounded-shape structures, they seem to have fractal morphology (perhaps PEG). In 3 perhaps you need dilute the sample. The black spots could be metallic spots (sample preparation??). Is this all the sampe sample??

3&4 are fatty acid-peptide-mPEG 5k and 7.5k
5 @ 6 are acetyl-N-GF3-peptide-mPEG 5k and 7,5k
What type of staining are you using? I would recommend uranyl acetate for a sample like this. It may be helpful to also analyze your samples by dynamic light scattering or AFM to corroborate your TEM results.
Thank you "Colin Calabrese ". I have used uranyl acetate for my samples. Also, I have done DLS. But How would DLS help in characterizing the geometrical shapes?
