Please help me in understanding the hairy root initiation condition. some paper suggest that after co cultivation in dark the explant without further bacterial infection outside is maintained on half strength MS media with cefatoxime and kept for incubation in 16h day/8h night condition for root initiation, whereas other papers maintain strict dark conditions throughout for root initiation. So please guide which protocol conditions are right for Hairy root culturing.
After co-cultivation, it is better to incubate the explants in dark for direct hairy root formation. For shoot formation from the hairy roots, can be kept in light. Please have a look of the paper

After cocultivation, it is not suitable to undergo root initiation directly. I belief, after cocultivation it should be kept for callus generation, and selection of transformed explants. Then shoot initiation from the callus, after that only, root initiation should be done.
