I am working with canine cerebrospinal fluid but my cytoslides do not contain good cells and the background seems dirty. I will be using these slides with IFF.
Concentrate the cells using regular centrifuge (500 - 650 rpm) for 3- 5 minutes. Then, resuspend in 0.5 ml saline. Finally, use Cyto•Fluid Fix™ and cytospin slides as the cerebrospinal fluid has sparse or no cells. Centrifugation at 650 rpm for 2 minutes. Stain after draying.
Did you ever consider filtraton instead of cytospinning? Filtration simplifies the procedure considerably with and has a very high yield. Take a look at www.vycap.com to find out if filtration is working for your purpose.
