Can anyone help me to optimize QQQ parameters for Dansyl-Estradiol...523...251?

I (attempted to) dansylate 100ng/mL d5-Estradiol using 200uL 100mM NaHCO3, pH=10.5, and 100uL of 1mg/mL dansyl chloride in Acetonitrile, incubation time = 20min, temp= 31.1C (targeted 30C). I am trying to tune/optimize QQQ parameters for the dansyl-d5-Estradiol, expected [M+H]+ = 511.4, all I'm seeing are 523, 251. Help!

I think maybe there is a problem with the derivatization step that you observe other ions than expected, and the process was not efficient or did not go correctly. Here is some application from Agilent for your compound, maybe try their parameters:
