Hello everyone,

I have a question regarding OT-1 stimulation and cytokine production analysis by FACS.

I have introduced a transgene in OT-1 mice that is supposed to lower the cytotoxicity of those cells. I need to confirm that. I am not studying memory cell development or its ability to differentiate into different subsets. I need something similar to an MLR. 

What would be the best method to measure IFN-g (the technique that should give me the "highest" levels of IFN-g secretion) ?

A reference would be highly appreciated for the details.
