I have got O.D. for estimating Ca from serum by spectrophotometer. Does anyone have molar extinction coefficient of Ca estimation or can give me values of standard conc. of Ca.?
Your question is not clear. In the medical laboratory, total Ca is usually measured in serum using the o-cresolphthaleine- or arsenazo-III-method. Both have distinguished color rections with different extinction values.
This was based on the method of precipitation by chloranilic acid (Cerioti, 1974). To 0.5 ml of the serum in a centrifuge tube was added 0.5 ml of chloranilic acid. This was
mixed thoroughly and centrifuged. The precipitate was washed in 3 ml of 50 % prapanol–water mixture, and later dissolved in 3.5 ml of citrate buffer (0.2 mol/l). The mixture was shaken on a “cyclomixer”, and the absorbance read at 530 nm against water as blank.
