Immediately after injection there is mobility impairment stretching and waist pinching. This reaction lasts for about 45 seconds and then the animal regains normal posture and behaviour.

45 sec is few time, but with some experience working with animals lab you can get some pain signal as postration of animal after the application of some product, like other signal of pain, for eg faster move when have irritation or ardency, rats runs around the cage. In the case of Hydroxyurea, this is a medicine for cancer therapy right? this is oral administration in human I hope so? and not sure, but any way, you must to chekec or see which it is the route of administration, because if no the same, ideally use the recommended route. There is some products that from the moment of application you can see a reaction pain or disconfort.

Thank Reynaldo, Yes, HU is a cancer drug that is given oral in human and comes with many warning that it is a skin irritant and to avoid contact. Some patients have symptoms of a burning sensation when on the medication. I have never encountered this before with other chemicals in my research so I was just startled by the response. Dear Marnie, I'm working with animal models of pain, and I can tell you that the first 45 sec after injection should not be taken into any kind of consideration. That reaction that you observed is the consequence of injection in peritoneum. And as is said if the compound has been registered to cause burning sensitization...there is your answer.
Thank you Nikola, Yes the transient pain is actue lasting 45sec to 1:35sec. This is my conclusion as well a burning sensation. This unexpected reaction has opened my eyes to the research on pain. What an interesting field of study.

I think you dose is too much decrease the amount, while if you need this such dose as experimental model, I prefer to prepare a mixture of it with Xylazin because it make rats more relax when you are doing experiment
