I have heard from Martin Chapman years ago, that IgE could not bind to Der p 1 after SDS-PAGE (with or without DTT). From my experience, I confirm this.
The native conformation in Der p 1 depends of disulfide bridges. Disruption of these bridges can alter the conformational epitopes of this allergen. For this reason, you must avoid the use of B-mercaptoethanol or heat. All this bring to the lost of IgE reactivity.
from our experience, Der p 1 can not be detected by IgE by SDS-PAGE whatever the treatment (heat, DTT or not). We succeed to detect it by rabbit polyclonal anti-Der p 1, after reducing condition (DTT), only.
