I am cultivating primary chondrocytes in DMEM-Media with varying glucose concentrations of 1 g/L (FG 0415) and 4,5 g/L (FG 0445) with the addition of initial 1% FCS, 1 % Penicillin and Streptomycin, 1 % amphotericin, 14,2 mM Ascorbic Acid and MEM non-essential amino acids (1:100). The cells were initially on 10 % FCS for adhesion and for reasons of stimulation culture reduced to 1 % FCS. After roughly 5 h one can observe crystal like formations in low and high glucose media. The cells were incubated at 37°C and 5% CO2 .
These are cell debris that are formed from collagen and gag aggregation. You can use 45 or 75 um cell strainer to remove them.
To reduce the number of debris and fiber before cell culture you filter your cells on cell-strainers. I use the one we put on the top 50mL tube. A second point, I think the cell density is a little low for choncdrocyte.
