Hello could anyone clarify my doubt here

I am working with invertebrates to assess the toxicity of chemicals.

Question 1

I exposed 25 animals to a toxicant (5 different doses) and my study period is 72 hour. on 48th hour 5 animals and 8 animals were dead in 4th and 5th doses respectively and all the remaining animals were dead by 72 hour. Now i wanted to calculate LC 50 value from the test. Here what should be my total number of animals? i have taken 25 animals initially but at the end of 72 hour only 20 and 18 animals were remained. So what should be the total no of animals 25 or 18 (5th dose)?

Question 2:

Is it necessary that all the experimental animals should die when calculating LC50 value?

Question 3?

I am setting my dose range in micromolar say 100, 120, 140, 160, 180 and my LC50 falls between 140 and 160 micromolar. For subsequent experiments like gene expression should i treat the animals with LC50 value or i can opt 160 micromolar value.
