I'm looking for a suitable tissue grinder to homogenize whole mite samples. I'm going to use these extracts as antigens for coating ELISA plates. Thank you!
Use Kauntes, glass to glass homeginizer, it will be best because of heat adjustment and no de-naturation of material occurs. A dounce homogenizer is the best way to get proteins extracted in solution. Basically, cell disruption is needed for the extraction of intracellular products. The method used may vary depending on the type of cell and its cell wall composition. Irrespective of the method used, the main objective of cell disruption must be effective and the method should not be too harsh so that the product recovered remains in its active form. There are different methods available for cell disruption. In recent trends, intracellular products which are of commercial value are released from microorganisms mainly by disrupting the cells by mechanical methods
If you are working with mite samples for protein extraction, then your sample size is perhaps small. I have used the Ultra-Turrax T10 Rotor Stator Homogenizer from IKA. Using this, you can basically extract proteins in a 1.5mL tube or a 10-15 mL tube. I have worked with house dust mites previously and this homogenizer has given me really good protein extraction yields. As Ravi Kant Upadhyay rightly mentioned, the dissipated heat may denature the proteins. Therefore, while using this homogenizer, always maintain the tubes on ice and homogenize in a pulsed manner.
