We have a mouse model where we see a decrease in total tau protein. We would like to verify if it is due to axonal degeneration or something else. Do you know of some good antibodies for specific markers of axons (other than tau protein). We've read that NFL is a good marker and we would like a good antibody that would work in IHC and Western. Apart of tau and NFL, is there any other marker specific of axons?

Hi Simon, your answer is not very helpful. You just referenced your company website and antibodies but do not show an antibody that is specific for axons. The staining from the NFH antibody show the soma and dendrites as well, it's not specific for axons.
Cher Fred, I can see that I'm not the only one with the problem. I think I found a solution. The mice where we see the decrease in tau after treatment are the hTau mice from Peter Davies and Karen Duff. They express human tau on a murine tau KO bajcground. Since the murine tau KO was done by inserting a GFP at the beginning of the tau sequence, they express GFP only in axons. So we'll do staining and WB for GFP and see what happens.
Big problem in IHC.
We have used anti-ZnT 3, a synaptic vesicle located zinc-transporter that labels hippocampal mossy fibers (https://www.sysy.com/products/znt3/facts-197003.php). Much better than Timm staining.

See Fig 8 (http://www.jneurosci.org/content/31/1/105.long). In our hands 1/2000 for IF. You can see the axonal vesicles. It is an indirect form, but you can also analyze trnasport. Good luck.
