Hello, I have a problem with dissolving chemical that from chemical library.
I want to test it for in mouse, so I need to dissolve it and make a high concentrated stock for mouse.
But it isn't soluble to PBS and DMSO or PEG mixtures even 80 C heating.
I asked what solvent would dissolve this product to chemical synthesis company,
they said "It is not soluble in water. Possible solvents for them : DMSO, acetonitrile, chloroform etc. To increase the solubility, you need to either warm up the solution slightly (up to 60-80 C), or add more solvent."
Despite of their advice, and it is safe for mouse, it dose not help me to dissolve chemical.
Can anyone helps me to solve this problem?
Solvent must be safe in mouse when do the per oral injection.
Did you try to dissolve it in a mixture of DMSO and PEG 400 and other solvent ?
It is difficult to help you without knowing the chemical that you are trying to dissolve and/or its chemical structure. A chemical formula and molecular weight might provide useful information if you can not reveal the chemical identity. One class of solvents that you did not mention are the alcohols such as methanol and ethanol, or even perhaps an emulsifier such as tween 80 or other tween.
According to your description it is clear that your compound is hydrophobic in nature, soluble in non polar solvents. For animal application water is a good solvent. In order to dissolve it in polar solvent like water you need a bio compatible emulsifier/surfactant at a concentration just above its CMC level.
Also you can try ethanol but methanol may be toxic for mice.

Thank you for your kindly recommendation. It's molecular weight is 390.4781
and chemical formula is C23H26N4O2.
I'm worried about alcohol is toxic to mice. And I'll try to dissolve it in Tween 20.

You can use a mixture of PEG 4000, N,N-dimethyl acetamide, dissolving it in PBS or normal saline to reduce the carrier concentration below the LD50 values in order to prove non-toxic to the control group. Another alternative is using DMSO with Cremophore EL, using Ethanol as diluent but keep the cremophore concentration below 15% otherwise the solution will precipitate.
