I am working on Alkaloids,i want to separate these alkaloids from plant sources,now i got crude extract,now my idea is to separate crude alkaloids from total crude.
I saw some paper said that do Acidification and basification process for separation of crude alkaloids.Here,my question is how to Acidification and basification process.
Alkaloids are a type of secondary metbolites that you can easily "separate" from other compounds in a crude extract. First you disolv your extract in DCM or CHCl3 and put the solution in a separation funnel. Then add HCl 1N to pass all alkaloids to the aqueous layer. You need to do three or more extractions to be sure all alkaloids were extracted from the organic solution. If your extract is not soluble in chlroform or DCM just try to disolve the crude extract as much as you can with the acid solution and then filter. To libearte the alkaloids you need to add NH3 o sodium carbonate solutions to the acids solutions until get a pH 10-11. Finally you recovery the alkaloids with DCM, CHCl3 or ethil ether.
