I just read a phD thesis, which is written in Chinese, the conclusion of this thesis is that, 0. 3 -0.5 M EMS treat seed for 8 h is the best LD50, it depends on the wheat variety, when I studied in Japan some years agao, I learn from a Japanese, you should optimize the condition by yourself based on your own material, anyway, I would suggest make a gradient from 0.3 to 0.5 M to get the best LD50 you need.
Each batch of EMS is slightly different than the one before, therefore a titration of kill curves should be performed to achieve an appropriate LD50 concentration for your particular wheat accession/cultivar/line. Not only will each EMS batch potency be different, but the response from different genotypes will also vary. Good luck =)
0.3 to 0.7 is being concluded in various articles with different temp as well as treatment duration but more specifically I am looking for spring wheat.,

@Keiko M Tuttle thanks for giving your opinion, but can you please elaborate what's your suggestion : is it that I have to use the same batch of EMS for every treatment, cant I use different batches of EMS belonging to same company. and can you please suggest me which solvent will be the best for preparing the stock and working solution.
