my value of sample is greater than that of standard and when I consider the formulas such as
(1) Percentage inhibition = (Abs Control – Abs Sample) x 100 / Abs control
(2) % Inhibition=100 − (A1-A2)/A0)*100)
(3) % Denaturation inhibition = (1−D/C) ×100%
Where D is the absorbance reading of the test sample, and C is the absorbance reading without test sample (negative control).
(4) PERCENTAGE INHIBITION = [100-(OPTICAL DENITY OF TEST SOLUTION – OPTICAL DENSITY OF PRODUCT CONTROL) ÷ (OPTICAL DENSITY OF TEST CONTROL)] ×100.
Which can be correct????? If I put control first ie, rewrite (3) as
% Denaturation inhibition = (1−C/D ×100% Getting % otherwise percentage is exceeding 100.
Maybe adjust your equation....
From:
PERCENTAGE INHIBITION = [100-(OPTICAL DENITY OF TEST SOLUTION – OPTICAL DENSITY OF PRODUCT CONTROL) ÷ (OPTICAL DENSITY OF TEST CONTROL)] ×100.
To: [ODControl - ODInhibitor/ODControl] x 100
Test your control against itself ([ODControl - ODControl/ODControl] x 100 = 100. Then repeat with real test article.
It should work.

If it is same control value, as you know it is equal to zero. So can you please clarify that whether it is ([OD test Control - OD product Control/OD --------------- Control] x 100 = 100
Sir, I am getting the OD values where absorbance of control (OD) is a less value compared to OD of inhibitor. So with this formula ( [ODControl - ODInhibitor/ODControl] x 100), I get a negative value. So I am doubtful about my procedure and also I am doubted that whether this absorbance value gives the amount protein which left intact without denaturation (should gives minimum 'OD' only in control) or amount of denatured protein (here "OD" should high for control). I attach files with the procedure which I follow. I hope you can give a twist in my work. Thank you...
