I am studying alpha amylase inhibitory activity of some plant extracts by using DNS color reagent. While performing bioassay I have observed that the solid form of enzyme after putting into 0.02 mM phosphate buffer (containing 0.006 mM NaCL) pH 6.9 does not showed clear solution. It indicate that it is not completely dissolved in the buffer. Is there any other way to prepare alpha amylase solution?

and also let me know that once we prepare the enzyme solution is it required to maintain cold temperature?
