While working with colloidal chitin, I am consistently getting negative values compare to blank in DNS assay. The only difference in blank and sample is the addition of colloidal chitin. The only reason for the negative value I can think of is perhaps the pellet attaches to and buries some of the sugar with it. I have run spectrum on blank cuvettes, changes the cuvettes used for blank with sample, and the experiments have been run a large number of times (>50). Also, the difference in values between blank and sample is significant.
