to calculate the extraction efficiency, you have to know the initial/real concentration of protein in barley grain. Then the extraction efficiency will be:

extraction efficiency (%)= proteins extracted/proteins real *100.

Agree with Aurora Silva but dilution factor must also be considered after extraction protocol. May be, this was the confusing thing for your calculation.
