I want to do a cell based assay. This assay contains 5 uL of my cells (Dissolved in DMEM medium) and 5 uL of compound (dissolved in DMED medium also).

Suppose I count the cells in my suspension (10 mL) =750000cells/mL.

And In the assay I want 5000cells/well (in the 5 uL). Total number of wells = 48 (corrected 48.5). So based on that total volume = 242.5 uL.

but when i calculate I used this formula; volumed needed from suspension = 48.5 *5000/750000 = 323 uL. Is that calculation right ?

This means I should take 323 uL from the 10 mL suspension and pipetter 5 uL into each well?
