I am running a co-culture experiment with HeLa (mammalian cells) and various bacteria (P. disiens, P. bivia, L. crispatus). I need to measure the viability of the HeLa cells following a co-incubation period. I know i can centrifuge and separate the bacteria and HeLa cells using centrifugation and other wash methods, however some of the bacteria will have invaded into the HeLa cells. I have been trying to find a viability assay that won't allow the bacteria to interfere with results. 

Calcein AM assay will invade live cells, so i think this could work, however does anyone know if bacteria would somehow interfere with this assay?? If anyone has any ideas on different viability assays that would be great.
