I am currently doing some experiments with caco-2 cells. I'm looking at barrier integrity and probiotic conditioned treatment. I'm just trying to see what passage number the cells need to be at for these kinds of experiments. I've scanned the literature and I get a range of between 20 - 50. Is that correct? Does anyone have any input?

Use of lower passage numbers in case of Caco-2 cells will serve as a valuable in vitro model for studying intestinal barrier integrity and the effects of probiotics on barrier function.
Using cells with a passage number below 20 is recommended because higher passage numbers can lead to changes in cell characteristics and differentiation.
Lower passage number will help ensure that the model remains representative of the intestinal epithelial barrier.
