I'm doing research on cellulose content in cereal leaf (transgenic plants). I have collected T0 seeds and for each line of T0, I planted 20 seeds to get 20 T1 plants which will be used for further experiment. Since segregation is involved, it will result in plants with the inserted gene and plants without the inserted gene among the 20 plants (T1). So, in this case, each of the T1 plants are considered to be genetically different. I will take only one plant (with inserted gene) from each line and use the leaf for different treatments before subjecting them for further tests (enzymatic and gene expression tests).

Questions:

1) Normally for any experiment, we need at least 3 replicates. So, since I only take sample from one plant per line, is it considered as biological or technical replicate?

2) If it is considered as technical replicate, for each treatment that i have, can I pool the leaf from that one plant, grind into powder and then divide them into 3 technical replicates?
