I stained human atrial sections (1 µm) with CST #2146 ß-tubulin (1:75) but I sometimes get a striated staining pattern.
Antigen retrieval was at pH 9. Blocking/permeabilisation with 0.1% Triton X and 5% BCS. Overnight antibody incubation. Abberior STAR secondary antibodies were used (1:200).
According the manufacturer: "In regard to isoform recognition, based on homology, our β-Tubulin Antibody #2146 should detect β-Tubulin isoforms A, B, and C. The antigen sequence has 96% homology with beta 2C, beta 4B, beta 5B, and beta 4A isoforms 1, 2, and 3, and 95% homology with beta 2C isoforms A and B. Therefore it is likely to detect these subtypes." But as far as I know, only ß-tubulin III shows striations in cardiac myocytes.
I am wondering if this is just due to damaged tissue, but costained proteins performed quite well. Has anyone had this experience? Thanks!
