I am wanting to start work on organotypic and primary cell cultures of post natal rat hippocampus and i am not sure which media promises good results, should i go with MEM+ FBS or DMEM+FBS or Neurobasal + B27 supplement, please suggest me.
hi...we do these hippocampal culturing everyday...so the best medium is NBM +B27+ glutamine supplement

thank you for the information and It would be a great favour if you could forward me the protocols and recipes:).
We used Neurobasal media for Organotypic cultures with B27 supplement and it worked fine for me. Initially we cultured them in NB and 12% FBS and after 48-72hrs we cultured them in serum free media. If you can grow the cultures without antibiotics it would give better results, but if in case its not possible then you could use Gentamycin and streptomycin we did but please check for the optimum concentration you could use.
Now coming to Primary cultures we got good results when we used RPMI + 10% FBS+ Gentamycin and streptomycin.
