If you have a microscope and hemocytometer in the field, suggest you use Randolph’s Stain to perform absolute eos count. It’s Phloxine B and Methylene Blue dyes. Eosinophils stain a majenta/purple color. Don’t recall the formula at the moment.
Which type of sample do you want to stain? For mouse bronchoalveolar lavage samples we successfully used Diff Quick (which is H&E based). For tissue samples (i.e. lung cuts), one can detect esinophils on H&E stained slides (not always easy, but works if the staining is done nicely) or with LUNA staining. There are some good papers out about other eosinophil-specific staining protocols (i.e. PMC2664312).
