I am currently working with BMMC and HMC-1.2 which are both floating mast cell lines. I want to run a time toxicity study (alamar blue or MTT) using different drugs and evaluate the % viability after short time frame (i.e. 5 min up to an hour). I was wondering if anyone had a protocol or a good approach for running time/concentration dependent toxicity studies with suspension cells. Specifically, after incubating the cells with the drug for a specific time, what is the best approach for removing the media and adding fresh media before incubating with alamar blue reagent? Is it necessary to remove the media containing the drug before adding the reagent? I am just worried that the constantly centrifuging the cells will interfere with the results.
