Hello,

We are currently working on an inflammatory model using a mouse air pouch and we are having some trouble with evaluating gene expression using this tissue. We have been using the Qiagen RNeasy plus micro kit for RNA extraction and our RNA yields have been extremely low from our air pouch (as low as 1.1ng/uL). Typically after collection we add tissue to RNAlater solution and store in -80. Our samples vary a lot in weight between 0.02g to 0.2g.

Does anyone have any experience working this with air pouch tissue? Is there a better method for doing RNA extraction and what could be the reason we are getting such low yields? Is there a specific homogenizer that works best with this type of tissue?
