I use BDNF immunofluorescent staining in brain and gut. In intestines, it works perfectly, and the staining looks specific according to the literature. However, same protocol with the same antibody applied to the brain doesn't work. I use the same blocking serum, same washing buffer, same secondary antibodies, same incubation at the same temperature,..

Do you have any ideas on how to overcome this problem? I was thinking about some permeabilization techniques or maybe antigen retrieval for frozen sections?
